Are there any guidelines-based on either formal design considerations or material considerations, which could be used to determine when to add a center rail to a raised panel door. Doors will be made of poplar and painted. Approximately 35″ long.

Hi wink ,
It really depends on the exact width and the material the panels are made of . Solid wood panels will move more than MDF as a rule .
We keep most cabinet doors under 23" when possible give er take a little.
Style's like Shaker or Mission may call out for the split panels , some finished refrigerator end panels I have made 32" deep with like 5 narrow panels .
So the main reason of caution is a wood panel say 20 " wide may shrink or move conservatively 3/8" or so , when you split it they each move half as much.
On paint grade Poplar type of cabinets I would advise the use of MDF for the panels , it comes in different density's the one that is almost like tempered makes great panel stock and paints well .
