I’m ready to attach two back panels to the upper section of my armoire. The panels are 15 x 42 – 1/4″ oak plywood. Should I use just wood screws or is it ok to put a spot of glue at the centre point of the top and bottom of each panel?

You don't say how they will be joined to the carcass -- in a rabbet? That's the way I often do this when I'm not being "fancy". Ply doesn't move much, so you don't need to worry too much about wood movement. I usually just nail (or staple) or screw such panels on. If they are in a slot, no need for any fastening. Just let 'em float.

Thanks Mike. Yes, they are in a rabbet.
I set my back panels in rabbets and use a few 1" drywall screws to hold it in place and keep the carcass square during the glue up. After the carcass is done, I can easily remove the back while I put in drawer glides or whatever and do my stain/finish. Being able to work from the back is very useful - lol.
When everything is done, I reinstall the back with narrow crown staples.
