I just made a new cross sled, 16 x 24″ using 3/4′ MDF. Should I seal the MDF with shellac or another sealer, to prevent it taking on moisture? My workshop is in my basement, heated in winter and I run a de-humidifier all summer so the relative humidity is under “reasonable” control.

Absolutely, yes! The MDF will attract dust and dirt and will begin to smell if left unsealed/finished.
Shellac is an ideal sealer. Sand the sled smooth to 180-220 grit. Use one dilute coat to seal the MDF. Let dry for an hour. Then level the raised fibers with 320 grit. Then finish with a 2# cut.

Shellac will work fine or if you want a longer lasting coating, use polyester resin and dilute it, laying it on with a cheap brush in a thin coat. Any high spots can be sanded off after it cures. I used poly resin for my bench that is used for mainly non-woodworking, like greasy/oily or paint/solvent jobs. If it gets too nasty, I wipe it down with whatever solvent or cleaner I want or is handy.
