I recently purchased a Jet 6″ jointer which has a 4″ (nominal) port for dust collection. Presently I only have a shop vac available for this purpose. Does anyone have experience adapting a 4″ port to a Home Depot 2 1/4″ hose/shop vac?

A dust collector with a true 4" inlet/hose would be the best option for your JET 6" jointer.
You can attach your shop vac with a Universal Dust Port Adapter. ($5.00 at Rockler http://www.rockler.com/ecom7/showdetl.cfm?&DID=6&offerings_ID=10472&ObjectGroup_ID=32&CATID=7
Rockler also sells a 4" -> 2.5" Reducer (part #89195), and a Shop Vac Adapter (part #34654).

Thanks Dan for the Rockler part suggestion. I will try the chip collection method suggested by Jeff to see how it works out. I will check into the Rockler part next time I am in their neighborhood which is about 15 miles away.

A jointer produces a lot of chips, but little dust. (If you're edge jointing, the shop vac should handle the job with no problems.) Depending on the size of your shop vac, you may have problems with the hose clogging.
Why not skip the vacuum all together and just catch the chips with a box or bag set at the chip chute? (I do the same thing and I have a dust collector. Jointer is on wheels, and it's easier to catch the chips than hook up a hose.)

I agree with Jeff, never seen a need to run a collector hose to the jointer. I built a box from 1/2" ply that sits on the floor at the chute. The top edge of one side is beveled ( side against the machine) so chips fall in easier. I removed the round plastic port and used it on another tool. Some woods like soft maple can clog the passage but this is rare and easily cleared with a stick. You'll know it's clogged when chips start sailing your direction.
It sounds like this would be a good place for one of those 5 gallon paint can pre-separator units, let it take the chips and the shop vac handle any dust.

The 5 gal paint can chip collector is the miniatire version of the 30 gal trash can collector. Both are basically lids which use 2.5" or 4" pipe as inlets and outlets. You can get them from pretty much any woodworking place, rockler, woodcraft, etc. The device is placed between the DC/shop vac and the source of the chips, and acts to collect the bigger stuff for easier emptying. I actually used the 30 gallon model and my dc650 to suck 1000 lbs of old dirty rockwool insulation out of my attic.
As I wrote Mark I will check this out at my local Rockler/Woodcraft store. My 8x10 shop begs for small solutions. Thanks for the information.
