Couple of question concerning making small keepsake box using veneers. Boxes will built with solid wood stock. None of the boxes will be bigger than 8″x 12″x 16″. 1-Do I need to use backer veneer for the interior.
2- Is titebond 3 ok for veneers.
3- Plan to preveneer all the stock than size pieces, so which is better, using pva glue and the clothes iron method or making some presses out of mdf.

I make a lot of boxes that size or smaller. The conventional wisdom is to use either plywood or MDF for the sides, top and bottom, and then veneer both the inside and outside faces. When I am going to line a box with fabric, I usually don't bother veneering the inside face. I've never had a problem with the unbalanced veneer, probably because the pieces are small and there's already limited movement in plywood and MDF. You might run into problems with solid wood parts, even if you veneer both sides.
PVA glue and an MDF press (or MDF with clamps) should work fine. I prefer a specialty veneer glue like Better Bond X-Press (https://www.veneersupplies.com/products/Better-Bond-X-Press-Veneer-Glue.html).
