My wife and I are going to put a window into a large pass through that connects our office and great room. The pass through is framed with 2×4’s and we plan to build a hardwood frame that will hold the glass.
The glass will be about 40″ x 72″ and 1/4″ thick.
Should I use 1.5″ thick stock or can I get away with 3/4″.

All the weight will be downward so the thickness of the sides and top is irrelevant. As long as the bottom is well supported by the framing it doesn't need to be all that heavy either, but it may look better if the sill is thicker.

Thanks. I think it will look good if the vertical members of the frame are thicker as well, given this is such a large window. Most of the rest of the frame will be trimmed out, so the thickness will not show.
